Metabolic changes and their implications on the epigenetic landscape during neurogenesis

The aim of this thesis is to reveal how metabolic switches and in particular how the intracellular location of metabolic enzymes influence the activity of epigenetic enzymes during neurogenesis.

Description: The differentiation of pluripotent stem cells to neurons is accompanied by several metabolic switches that have broad implications for cellular development. The aim of this thesis is to reveal how metabolic switches and in particular how the intracellular location of metabolic enzymes influence the activity of epigenetic enzymes during neurogenesis. The thesis involves differentiation of iPSCs to neurons, the generation of organellar maps through LC-MS/MS based proteome profiling at different stages of differentiation, the characterization of the cellular metabolome by isotope tracing experiments and the analysis of the epigenetic landscape on the level of DNA modifications and post-translational histone modifications.
